脳梗塞の悪化抑える物質、大阪大など発見　新薬開発の可能性も

大阪大などの研究グループは5月19日、脳梗塞（こうそく）を発症したときに脳細胞の損傷範囲が広がるのを抑えるたんぱく質をマウスを使った実験で突き止めたと発表した。論文は近く、アメリカ科学アカデミー紀要電子版に掲載される。まひや意識障害など、脳梗塞の後遺症を抑える新薬の開発につながる可能性がある。時事ドットコムなどが報じた。

研究グループは、免疫を調節している「RANKL」と呼ばれるタンパク質に注目。脳梗塞を起こしたマウスにRANKLを投与すると、その後の炎症が約40％抑えられ、RANKLの働きを阻害するOPGというタンパク質を取り除いた場合には60～70％抑制されるのを確認した。

(中略)大阪大大学院連合小児発達学研究科の島村宗尚寄付講座准教授は「有望な治療薬になる可能性があるが、RANKLは骨粗しょう症を悪化させる原因物質でもあるため、さらなる検討が必要」と話している。

脳梗塞による死亡者数は日本で年間7万人を超える。介護が必要となる最大の原因疾患とされ、悪化を食い止めるには、発症後数日以内に起きる炎症の抑制が重要となる。
